DNA Sequencing and Microsatellite Analysis
NEW USERS
To use the CG Lab you will need to meet with the laboratory director for a brief interview about your project and experience, register (so we have contact info. and a billing address), and then set up a time for an orientation to the lab (where things are, approved protocols, safety training, etc.). After that, you keep track of any lab materials used (on a monthly charge form) and turn in your list at the end of each month. Sequencing activities are noted on the log next to the sequencer, special orders (of chemicals, special consumables, etc.), primers, and BigDye are also noted, and all charges are included on an itemized monthly bill given to you or sent to the address you specify.
CONTRACT SEQUENCING SERVICE
We are happy to process a few samples at a time for projects that just need an occasional few sequences, e.g. for clone confirmation. Small numbers of samples submitted for sequencing are usually finished within a week. We try not to do contract sequencing for large projects, and instead train students and post-docs to do the work themselves. As a special service, we occasionally will do a few hundred sequences for "external" users (or students who can't do the sequencing part of their project themselves).

and tracks lanes but user completes analysis

microsat charges include: acrylamide, ROX or TAMRA size standard
SUBMITTING MATERIALS FOR SEQUENCING
Please submit a SEQUENCING REQUEST FORM, purified DNA (e.g. Qiagen-cleaned PCR product or mini-prepped cloned insert) that is at least roughly quantified, and primer/s (unless it is one of the standard T3/T7/M13rev/M13-20 etc. cloning primers, which we have). For cycle sequencing PCR product we need around 50 ng per kb of product per direction, for plasmid preps we need at least 200 ng per kb of insert per direction (e.g. for a 2 kb insert, 400 ngx2 for both directions). Clearly labelled tubes and fully descriptive sample identifiers and quantification is helpful.
